摘要
探讨连翘酯苷对人工感染传染性支气管炎病毒雏鸡的肺组织JAK/STAT信号通路的调节作用,分别设连翘酯苷预防组高、中、低和治疗组高、中、低6个剂量组,并设利巴韦林阳性对照组、空白对照组和病毒对照组。提取雏鸡肺组织总RNA,用相对荧光定量RT-PCR的方法测定雏鸡体内干扰素-α,JAK,STAT-1mRNA的相对表达量。结果表明连翘酯苷预防组高剂量100 mg/kg、中剂量50 mg/kg和治疗组中剂量100 mg/kg对雏鸡体内干扰素-α的mRNA水平表达有诱生作用(P<0.01),治疗组中剂量显著上调JAK及STAT-1的表达,与JAK/STAT信号通路一些因子的上调相关,其机制部分与肺组织的JAK/STAT信号通路激活和相关信号的表达有关,这是连翘酯苷发生抗病毒作用的机制之一。
To investigate the effect of forsythiaside A on regulation of JAK/STAT signal transduction in the lung from the chicken infected by infectious bronchitis virus,the chicken were divided into prenvention groups of high,medium and low-dose and therapy of high,medium and low-dose which were treated by forsythoside A respectively,the control group of ribavirin and virus control group.Total RNA from lung were extracted in each group.The relative expression of IFN-α、JAK and STAT-1 were detected by relative Real-time fluorescence quantitative PCR.Forsythiaside A of prenvention group of high-dose 100 mg/kg、medium-dose 50 mg/kg and therapy group of medium-dose 100 mg/kg could induce IFN-α expression in chicken(P0.01),therapy group of medium-dose increased the expression of JAK and STAT-1,which was relevant to the high expression of some signals in JAK/STAT signal transduction.Forsythiaside A inhibited the activation of JAK/STAT signal transduction and the relevant signals expression in the lung from chicken,which was one of the mechanism of anti-virus effect of forsythiaside A.
