脂肪来源干细胞移植大鼠新鲜软组织创面后的效应和分布

The effect and distribution of adipose-derived stem cells transplanted to rat soft tissue wound

摘要 目的研究脂肪来源干细胞(ADSCs)直接移植于新鲜创面后所产生的效应及其分布特点。方法取大鼠腹股沟脂肪,经酶消化法提取ADSCs后培养传代,构建新鲜软组织创面动物模型。将DiI标记的ADSCs以点状注射方式移植到新鲜创面组织内,观察创面愈合情况。术后24d,切取愈合组织行石蜡包埋和常规HE染色;术后48d,再切取愈合组织行快速冰冻切片,直接在荧光显微镜下观察。结果细胞注入创面组与对照组比较,创面愈合时间平均缩短1~2d;术后24d,与对照组比较,细胞注入创面组的愈合组织内有更多的腺样结构及血管形成;术后48d,荧光显微镜下仍可见大量ADSCs在创面组织内停留,并呈现分层富集的现象,自身对照组见少量散在的荧光。结论ADSCs有利于创面愈合,在创伤组织内有一定的迁移能力,分布有一定的规律性。 Objective To investigate the effect and distribution of adlpose-denved stem cells (ADSCs) which had been transplanted to fresh wound. Methods Fat from inguen of SD rats was harvested, the ADSCs were extracted by enzyme digestion, and then cultured in vitro for passage. The model of fresh soft tissue wound was established. The ADSCs labeled with DiI were transplanted into the fresh soft tissue wound by multipoint injection. The healing of the wound was observed. The healed tissue harvested at 24 days was performed the paraffin embedding and the routine HE staining; those harvested at 48 days was performed rapid frozen section and then fluorescence microscope was used to observe the results. Results Compared with the control group, the healing time was less 1 to 2 days then that in the experimental group; there was more gland-like structures and vascularization at 24 days postoperatively; generous transplanted ADSCs with delamination and concentra- tion were observed in the wound under fluorescence microscopy at 48 days postoperatively. There was a little fluorescence scattering in the control group. Conclusion Locally injected ADSCs with ability of transfer could survived and be helpful to fresh wound healing and its distribution follows the certain regularity.
