摘要
目的 研究一种新型的缺血再灌注损伤皮瓣的动物模型。方法在解剖研究的基础上,于10周龄雄性Fisher大鼠背部制备对称性皮瓣,形成不同缺血时间的缺血再灌注损伤模型,进行两侧皮瓣存活率、微小血管成像评估及组织学检查。结果缺血6h的大鼠背部对称性皮瓣缺血再灌注损伤模型,其左右两侧皮瓣存活率、平均微小血管数及组织学检查差异均无统计学意义(P〉0.1)。结论以旋髂深血管为蒂的大鼠背部对称性岛状皮瓣(缺血6h)是一种较好的缺血再灌注损伤皮瓣模型,适用于药物、细胞及细胞因子等对缺血再灌注损伤的实验研究。
Objective To investigate a new flap model for ischemia/reperfusion injury. Methods Based on the anatomic research, paired rectangular island skin flaps were elevated on the dorsum of 10 weeks old Fisher rats, and made the flap model for ischemia/repeffusion injury with different ischemia time. Evaluation of flap survival, microangiography and histological examination were carried out seven days later. Results There were no significant difference between the left and right flaps of mean flap survival area, average number of vessels and vessel counting in HE staining after ischemia/repeffusion for 6 hours ( P 〉 0. 1 ). Conclusion Rat dorsal paired island skin flaps (ischemia time 6 hours) was a precise model for isehemia/reperfusion injury, this model will be especially useful for detecting the effect of angiogenic agents such as pharmacologic drugs, cells, and cytokines.
