摘要
目的比较金钠多与艾司洛尔对缺氧联合血管紧张素2(Ang-2)所致血管内皮细胞分泌内皮素(ET)的保护作用。方法将培养的血管内皮细胞分为四组:A组缺氧前加入10-5mol/L的Ang-2;B组在A组基础上加入10 mg/L的金钠多;C组在A组基础上加入5 mg/L的艾司洛尔;D组正常培养作对照。用放射免疫法测定作用前、作用后0.5、6、24 h各组ET含量。结果与D组比较,缺氧后0.5、62、4 h三个时间点A组的ET含量均增高(P<0.01),B、C两组ET比A组降低(P<0.01)。用药后0.5 h,B组ET比C组降低(P<0.05),用药后6 h和24 h更明显(P<0.01)。结论金钠多和艾司洛尔对缺氧联合Ang-2对血管内皮细胞的损害均有保护作用,金钠多的保护作用更持久。
Objective To compare the protective effects of ginaton and esmolol on vascular endothelial cells secreting endothelin in hypoxia combined with angiopoietin-2(Ang-2).MethodsVascular endothelial cells were divided into 4 groups of A(treated by hypoxia and Ang-2),B(given additional ginaton 10 mg/L before hypoxia),C(given additional esmolol 5 mg/L before hypoxia) and D(normal controls).The changes of endothelin expression were measured with radioimmunoassay before and at 0.5,6,24 h after treatment,respectively.Results The expression levels of endothelin at 0.5,6,24 h after treatment were higher in group A than those in group D(P0.01),which were lower in groups of B and C than those in group A(P0.01).Compared to group C,the expression levels of endothelin in group B were lower at 0.5 h(P0.05),6 h and 24 h(P0.01) after treatment.Conclusion Ginaton and esmolol have the protective effects on the injury of vascular endothelial cells induced by hypoxia combined with Ang-2,in which the effect of ginaton is longer than that of esmolol.
