摘要 目的研究硫酸氨基葡萄糖对软骨细胞株ATDC5增殖分化及细胞外基质基因表达的影响。方法将软骨细胞株ATDC5分为硫酸氨基葡萄糖组(A组)和空白对照组(B组)。阿利新蓝染色检测蛋白多糖(GAG)含量,碱性磷酸酶染色(ALP)检测碱性磷酸酶活性,逆转录-聚合酶链反应检测Col2及Smad2、Runx2等。结果硫酸氨基葡萄糖抑制了碱性磷酸酶的活性及Smad2和Runx2基因的表达,促进了GAG的合成与Col2基因的表达。结论硫酸氨基葡萄糖不仅促进软骨基质的合成,同时也影响软骨细胞株ATDC5分化。 Objective To investigate the effects of glucosamine(GlcN) on chondrocyte line ATDC5 proliferation and matrix gene expression.Methods ATDC5 cells were divided into group A(treated with glucosamine) and B(blank control).Alkaline phosphatase staining and alcian blue staining were used to determine the content of GAG and activity of alkaline phosphatase.RT-PCR was used to detect Col2,Smad2 and Runx2.Results Notably,glucosamine enhanced cartilage specific matrix components,collagen type II,inhabitant runx2 and Smad2.Conclusion Glucosamine sulfate not only promotes the synthesis of cartilage matrix,but also affects the cartilage cells ATDC5 differentiation.
