摘要
目的研究血管内皮生长因子(VEGF)在肺挫伤(Pc)肺水代谢中的作用。方法30只家兔随机均分为5组,每组6只:PC后2、4、6、8h观察组(A1、A2、A3、A4组)及正常对照组(c组)。自由落体撞击装置建立PC模型;免疫组化法和RT-PCR分别检测肺组织中VEGF蛋白和VEGFmRNA表达;称重法观察肺水参数的变化。结果与C组相比,PC各观察组肺水参数均明显增加(P〈o.01)。A1、A3及A4组VEGF蛋白及其mRNA表达较C组增加,高峰出现在PC后6~8h(P%0.01)。VEGFmRNA表达与氧分压呈负相关(P〈o.01)。结论PC后缺氧等因素通过上调VEGF表达促进肺水肿的形成。
Objective To investigate the effect of vascular endothelial growth factor(VEGF) on the pneum-fluid metabolism of pulmonary contusion (PC). Methods Thirty rabbits were randomly and equally divided into five groups of A1(2 h after PC) ,A2(4 h after PC) ,A3(6 h after PC) ,A4(8 h after PC) and C(normal control). Free fall hitting was used to establish the rats PC model. The expressions of VEGF protein and VEGF mRNA were detected by immunohistochemistry and RT-PCR, respectively. The changes of parameter of pneum-fluid metabolism were measured by weighting method. Results Compared with group C, parameters of pneum-fluid metabolism were significantly increased in groups of A1, A2, A3 and A4 (P〈0. 01). The expressions of VEGF protein and VEGF mRNA in groups of A1, A3 and A4 were higher than those in group C, which peaked at 6-8 h after PC(P〈0. 01). The expression of VEGF mRNA was negatively correlated with partial pressure of oxygen (r=-0.79,P〈0. 01). Conclusion After PC, hypoxia and other factors promote the ~ogression of pulmonary edema via up-regulating the expression of VEGF.
