12份不同地理居群野菊的遗传多样性分析 被引量:17

摘要 结合形态学性状和分子标记对12份不同地理居群野菊进行了遗传多样性研究。结果表明:不同地理居群野菊各形态性状存在不同程度的变异(14.06%~67.50%),叶柄长、叶长/叶柄长、花序径、舌状花长、舌状花宽和舌状花长/舌状花宽可能是造成野菊不同地理居群间表型差异的主要因素。25个ISSR引物共扩增到246条扩增带,其中229条呈多态性,多态性带比率平均为92.5%;29对SRAP引物组合共产生651条扩增带,其中638条呈多态性,多态性带比率平均为98.0%,从分子水平揭示了野菊丰富的遗传多样性。12个居群间的Nei's遗传距离分布在0.211 7~0.494 9之间;聚类分析表明,南京野菊和四川野菊遗传关系较近,武夷山野菊独立成一组,与其他地理居群野菊遗传关系较远,不同地理居群野菊的遗传变异与地理分布相关性不明显。 Morphological characteristics and molecular markers were used to study the genetic diversity among 12 populations of Dendranthema indicum.The results showed that morphology characteristics had considerable variances(14.06%-67.50%)among geographical populations.The variations of morphological characters of the D.indicum may be caused mainly by the leafstalk length,the leaf length/leafstalk length,the inflorescence diameter,the ray floret length,the ray floret width,and the ray floret length/ray floret width.ISSR fingerprinting amplified by 25 ISSR primers revealed a total number of 246 unambiguous bands,of which 229 ones were polymorphic and the polymorphism frequency was 92.5%.And SRAP fingerprinting amplified by 29 SRAP primer pairs revealed a total number of 651 unambiguous bands,of which 638 ones were polymorphic and the polymorphism frequency was 98.0%,which indicated a rich genetic diversity of D.indicum at the molecular level.The genetic distance ranged from 0.211 7-0.494 9.The UPGMA dendrogram suggested that populations of Chengdu Sichuan and Nanjing Jiangsu had the highest genetic relationship,while,the populations of them and Wuyishan Fujian had a relatively low genetic relationship,and there was no significant relationship between the geographical distribution and genetic diversity of D.indicum.
