川芎嗪联合大鼠骨髓间充质干细胞治疗大鼠脑挫伤实验研究 被引量:1

Study of TMP combined rat bone marrow mesenchymal stem cells in treatment of rats with brain contusion

摘要 目的观察川芎嗪联合大鼠骨髓间充质干细胞治疗大鼠脑挫伤的实验方法以及效果。方法选择试验用脑挫伤wistar大鼠,采用Feeney自由落体法,复制大鼠脑挫伤动物模型,将这些动物模型随机分为空白对照组,假手术组,川芎嗪治疗组,间充质干细胞治疗组和川芎嗪联合间充质干细胞治疗组5组实验,使用微量进样器打在损伤灶周围,第24h和第72h取脑切片进行abc免疫组织化学染色。采用图象分析技术统计神经元特异性烯醇化酶(NSE)、胶质纤维酸性蛋白(GFAP)的神经元数密度。结果3组治疗组的NSE和GFAP细胞数目高于假手术组和空白对照组(P<0.05),72h组明显高于24h治疗组(P<0.05)。结论川芎嗪对脑挫伤的大鼠海马周围皮层神经元有保护作用,其机制可能部分与半暗带Bcl-2表达有关。 Objective To observe the experimental methods and results of Ligustrazine combined rat bone marrow mesenchymal stem cells to treat rats with brain combined rat.Methods For testing brain contusion wistar rats were Fenne's free-fall method,rat brain contusion animal models,these animal models were randomly divided into control group,sham operation group,TMP treatment group,mesenchymal stem cell therapy group and TMP combined mesenchymal stem cell therapy group of five experiments,the use of micro injector to play around in the kitchen injury,24h and 72h abc of brain biopsies by immunohistochemistry.Statistical image analysis technique used neuron-specific enolase(NSE),glial fibrillary acidic protein(GFAP) density of neurons.Results Three treatment groups the number of NSE and GFAP cells compared with the sham group and control group(P0.05),72 h was significantly higher than that of the 24h treatment group(P0.05);Conlusions TMP on the surrounding brain contusion in rat cortical neurons of the hippocampus has a protective effect,which may be part of the penumbra with the Bcl-2 expression.
