摘要
目的探讨巴戟天多糖对实验性抑郁症大鼠模型氧化应激损伤及认知行为的影响。方法采用水沉纯提法提取巴戟天多糖,选择24只SD大鼠随机分为对照组、模型组及干预组,模型组及干预组建立实验性抑郁症大鼠模型,干预组给予巴戟天多糖干预,采用T迷宫实验观察三组大鼠认知行为变化,采用比色法检测三组大鼠血清MDA及SOD水平,采用荧光免疫法检测三组大鼠脑组织海马区神经元数目,比较三组大鼠T迷宫实验错误次数、血清MDA及SOD水平以及海马区神经元数目差异。结果 干预组T迷宫错误次数低于模型组、高于对照组,海马区神经元数目低于模型组,高于对照组,血清MDA水平低于模型组,高于对照组,血清SOD水平高于模型组及对照组。结论 巴戟天多糖能够减轻抑郁症大鼠体内氧化应激反应,减轻海马区神经元损伤,改善实验性抑郁症大鼠认知行为障碍。
Objective To investigate the effect of polysaccharides from morinda offcinalis (MOP) on the oxidative stress and congi- five-behaviors in experimental depression model of rats. Methods MOP was abstracted with water extraction and alcohol precipitation, 24 SD rats were randomly divided into control groups,model groups and intervention groups,depression model of rats were make,rats in intervention groups were interfered with MOP,change of congitive-behaviors were observed with T-type labyrinth test and number of nerve cell in hippocampus were detecting with fluorescence immunity,malondialdehyde(MDA) and superoxide dismutase level(SOD) in serum were detecting, error number of T-type labyrinth test,number of nerve cell in hippocampus and level of MDA and SOD were contrast. Results Mistake number of T-type labyrinth test was higher in intervention groups than in control groups,and was lower than in model groups,number of nerve cell in hippocampus was higher in intervention groups than in control groups,and was lower than in model groups,level of MDA in serum was higher in intervention groups than in control groups,and was lower than in model groups, level of MDA and SOD was higher in intervention groups than in control groups and in model groups. Conclusion MOP could release oxidative stress reaction and inhury of nerve cell in hippocampus,improve inprove the disorder of congitive-behaviors in an experimental depression model of rats.
