摘要
目的研究苦瓜蛋白对卵巢癌COC1细胞增殖与凋亡的影响。方法以梯度浓度苦瓜蛋白(0、10、15、20μg/mL)处理COC1细胞24h,以10μg/mL苦瓜蛋白处理COC1细胞不同时间(0、12、24、48h),行台盼蓝染色、流式细胞术、MTT法分别检测细胞存活率、细胞凋亡率、细胞增殖抑制率;10μg/mL苦瓜蛋白处理COC1细胞作用不同时间后提取细胞总蛋白,Westernblot检测多聚二磷酸腺苷核糖聚合酶(PARP)蛋白的表达。结果 苦瓜蛋白可以明显诱导COC1细胞凋亡,以20μg/mL苦瓜蛋白处理组细胞凋亡率最高;苦瓜蛋白对COC1细胞增殖有明显的抑制作用。苦瓜蛋白处理COC1细胞24h后,PARP表达明显上调。结论 苦瓜蛋白可以诱导COC1细胞发生凋亡,抑制细胞增殖,其作用机制与PARP有关。
Objective To investigate the effects of momordicin on proliferation and apoptosis of human oophoroma COC1 cells. Methods Trypan blue staining, cytometry and MTr assay were applied to detect survival, apoptosis and proliferation status of COC1 cells;Western blot was applied to determine expression of apoptosis-related gene PARP. Results Results of trypan blue staining, cytometry and MTT assay showed that momordicin induced apoptosis of COC1 ,and the highest apoptosis rate was in the group treated by 20 μg/mL momordicin; Furthermore, momordicin significantly inhibited the cell proliferation. Western blot results showed the up-regulation of PARP fragment in momordicin-induced group. Conclusion Momordicin could promote apoptosis and inhibit proliferation of COC1 cells and the potential mechanism may be involved with PARP.
