摘要
以豫杂一号泡桐丛枝病幼苗为材料,研究了硫酸二甲酯处理对其幼苗形态和基因组DNA的SSR扩增位点的影响。结果表明,质量浓度为15 mg.L-1的硫酸二甲酯处理3 h幼芽形成的幼苗仍呈现丛枝病幼苗症状,而质量浓度等于15 mg.L-1、处理5 h或大于25 mg.L-1、处理超过3 h的幼芽形成的幼苗形态上均呈现健康状态,但15 mg.L-1硫酸二甲酯处理5 h的幼苗内仍有植原体存在。此外,豫杂一号泡桐丛枝病幼苗、健康幼苗、15、125 mg.L-1硫酸二甲酯处理幼苗的DNA在SSR水平上扩增位点相同。
An experiment was conducted to study the effects of dimethyl sulphonate on the morphological changes of Paulownia tomentosa × P.fortune seedlings with witches' broom and genomic DNA-derived SSR loci.Results demonstrated that the seedlings treated with 15mg·L-1 of dimethyl sulphonate for 3h remained diseased symptom and those treated with 15mg·L-1 for 5h or with more than 25mg·L-1 for longer than 3h stayed healthy.But the phytoplasma was detected in the seedlings treated with 15mg·L-1 for 5h with net-PCR.Moreover,the genomic DNA basal sites of the diseased seedlings,the healthy and the ones treated with 15mg·L-1 or 125mg·L-1 dimethyl sulphonate were the same at SSR level.
