TRAIL对人喉癌Hep-2细胞生长及凋亡的影响

摘要 目的:研究TRAIL对人喉癌Hep-2细胞生长及凋亡的影响,并探索其相关机制。方法:用MTT法和流式细胞仪检测不同浓度TRAIL对喉癌细胞Hep-2,成纤维细胞MRC-5生长及凋亡率的影响,用western blotting对比两种细胞DR-4,DR-5的表达情况。检测Hep-2细胞TRAIL干预前后caspase-8,caspase-3,caspase-9表达的变化。结果:随着TRAIL浓度的升高,Hep-2细胞存活率逐渐降低,而凋亡率逐渐提高。MRC-5细胞对TRAIL不敏感。Western blotting检测发现Hep-2细胞DR-4、DR-5表达明显高于MRC-5。TRAIL干预Hep-2细胞后caspase-8、caspase-3、caspase-9活化分解明显增多。结论:喉癌细胞Hep-2由于DR-4、DR-5的高表达,对TRAIL促凋亡作用具有特异敏感性,其促凋亡作用是通过激活线粒体依赖性通路实现的。
