摘要
目的:探讨透明质酸介导运动的受体(receptor for hyaluronan-mediated motility,RHAMM)和ERK2(extracellular signal-regulated kinase)在结肠癌组织中表达及其与临床病理因素的关系,评估其在结肠癌诊断及预后中的价值。方法:收集我院结肠癌患者标本30例及其相应癌旁组织,通过real-timePCR对RHAMM和ERK2进行检测。结果:RHAMM和ERK2在结肠癌组织较癌旁组织表达均明显升高(P<0.001),且两者表达在低分化、中分化和高分化3组组间差异无显著性(P=0.222和P=0.173),但都与是否发生淋巴结转移有关(P=0.01和P<0.001);两者表达具有一定相关性(P=0.009,r=0.465)。结论:结肠癌中的RHAMM和ERK2表达都高于正常组织,且两者表达有相关性,RHAMM可能通过Ras-ERK途径参与了结肠癌的发生、进展及转移,RHAMM和ERK2可作为患者诊断和预后的参考指标之一。
Objective To investigate the expressions of receptor for hyaluronan-mediated motility(RHAMM)and extracellular signal-regulated kinase(ERK2)and their correlations with clinical pathological factors of colon cancer,and to evaluate their value in diagnosis and prognosis of colon cancer.Methods Real-time PCR was used to detect the expressions of RHAMM and ERK2 in 30 colon cancer tissues and 30 matched tumor-adjacent samples.Results The expressions of RHAMM and ERK2 in tumor tissues were significantly higher than those in tumor-adjacent samples(P 0.001),and were not associated with tumor differentiation(P = 0.222 and P = 0.173,respectively),but were associated with lymph node metastasis(P = 0.01 and P 0.001,respectively).There was significant correlation between the expressions of RHAMM and ERK2(r = 0.465,P = 0.009).Conclusion The expressions of RHAMM and ERK2 are higher in colon cancer than those in tumor-adjacent samples,and they are correlated to each other.Which suggest that RHAMM may involve in the pathogenesis and metastasis of colon cancer through Ras-ERK pathway.Expressions of RHAMM and ERK2 may be helpful in the diagnosis and prognosis of colon cancer.
