摘要
目的建立一种简便、实用、稳定、可靠的慢性锰中毒动物模型。方法 SD大鼠腹腔每天注射15 mg/kgMnC l2.4H2O 12周,同时设立对照组,观察锥体外系组织(纹状体)组织学变化及大鼠肌张力等指标改变;并对脑组织取样测定锰(Mn2+)、多巴胺水平变化。结果染毒结束后SD大鼠出现确定的肌张力增高、平衡失调;电镜观察可见神经细胞变性和神经纤维脱髓鞘改变;大鼠脑部锰含量增高,锥体外系组织(纹状体)的锰含量增加4倍以上,多巴胺合成减少,符合慢性锰中毒的损害定位。结论用此方法可以制备理想的慢性锰中毒的动物模型。
Objective To establish a simple,practical,stable and reliable animal model of chronic manganism.Methods SD rats were treated with MnCl2·4H2O(15 mg/kg)by intraperitoneal injection every day for 12 weeks,at the same time,normal control group was set up.The histological changes in extrapyramidal system(striatum)and muscular tension of SD rats were observed;the levels of Mn(Mn2+)and dopamine in SD rats brain tissues were detected.Results The muscular tension increased and a certain balance unsteadiness was found in SD rats after exposure,nerve cells degeneration and nerve fibre demyelination were observed under electronic microscope;the content of Mn increased in brain of SD rat,the content of Mn2+ in extrapyramidal system increased more than four times,dopamine synthesis decreased,which conformed to chronic manganism.Conclusion This method can be used to establish an ideal model for chronic manganism.
