摘要
目的:探讨姜黄煎对大鼠血管损伤后血小板内皮细胞黏附分子PECAM-1与hif-1α表达影响。方法:将144只大鼠随机分成4组,姜黄煎干预组,生理盐水干预组,假手术组和正常组(对照),每组36只。模拟PCI术,采用组织形态学镜下观察急性损伤后大鼠血管内膜结构的变化,电镜下观察细胞凋亡情况,ELISA法检测血清中PECAM-1与hif-1α含量。结果:(1)姜黄煎组、生理盐水组PECAM-1与hif-1α含量与假手术组、正常组比较差异明显(P〈0.05);(2)假手术组PECAM-1与hif-1α水平含量接近于正常对照组,差异不明显(P〉0.05);(3)姜黄煎组与生理盐水干预组、假手术组比较有明显差异(P〈0.05),与正常对照组比较差异不明显(P〉0.05);(4)通过血管内皮形态,可见大鼠动脉球囊损伤后内膜增生在术后早期(37天)就已达到高峰,姜黄煎组可抑制内膜增厚。结论:姜黄煎可上调大鼠hif-1α蛋白表达,使PECAM-1的蛋白表达明显下降,说明hif-1α过量表达,PECAM-1/hif-1α的比值减小,能抑制内皮细胞过度凋亡,有助于评估血管损后凋亡因子对血管内皮功能和结构的影响。
Objective:To investigate the effect of turmeric fried platelet endothelial cell adhesion molecule PECAM-1 and hif-1α expression in vascular injury rat.Methods: A total of 144 rats were randomly allocated to four groups,36 of each group,including fried turmeric intervention group,saline treated group,sham group and normal group(control).Analog PCI was applied and morphology were observed in rats after acute injury intimal changes in the structure.Apoptosis and serum content of PECAM-1 and hif-1α were detected by Electron microscope and ELISA,respectively.Results:(1)Levels of PECAM-1 and hif-1α,in turmeric fried group,saline group,sham group,were significantly different(P〈0.05)compared with the control group.(2)There were no significant differences(P〉0.05)between sham group and control group.(3)The concentration of PECAM-1 and hif-1α in turmeric fried Group were significantly different(P〈0.05)with the saline group,sham group,and showed no significant differences(P〉0.05)compared with the control group.(4)Changes of vascular endothelial morphological had reached a peak after balloon injury of rat carotid artery intimal hyperplasia in the early postoperative period(37days),turmeric fried Group inhibit intimal thickening.Conclusion: The expression of hif-1α was up-regulated by turmeric fried while PECAM-1 significantly decreased,indicating over expression of hif-1α.Ratio of PECAM-1/hif-1α decreases and excessive apoptosis of endothelial cells could inhibit.It may help to assess the effect of vascular injury induced factor on vascular endothelial function and structure.
