摘要
目的利用量子点(QD)优良的光学性质对人舌癌Tca8113细胞内bax进行特异性荧光标记。方法利用QD545nm通过间接免疫荧光法,在激光共聚焦显微镜下观测人舌癌Tca8113细胞内bax的表达,并激光连续照射1 h,用激光共聚焦显微镜自带软件Leica Confocal Software测量量子点QD545nm的荧光信号强度。结果激光共聚焦显微镜下可见人舌癌Tca8113细胞内bax明显表达,且激光连续照射1 h,QD545nm标记的荧光未见明显衰减,而FITC标记的荧光1 h内已基本淬灭。结论量子点荧光标记技术能对细胞内bax进行标记。
Objective Utilizing the excellent optic characteristic of quantum dots(QD) to fluorescence labeling on bax in human tongue cancer Tca8113 cells.Methods Quantum dots(QD545nm) and fluorescein isothiocyanate were respectively applied to tag Tca8113 strains with indirect immunofluorescence,the expression of bax in Tca8113 tongue cancer cells was observed under confocal laser microscopy and after continuous irradiation of laser for 1 hr as well.Results With laser scanning microscope QD545nm quantum dots marked bax expression was clearly seen in human tongue cancer cells Tca8113.After one hour's continuous laser exposure,there was no obvious declining of fluorescent markers.Conclusion Quantum dot marking fluoresent technology can tag bax in cancer cells
