益肾活血方对早期糖尿病肾病大鼠作用机制研究 被引量:6

摘要 目的:探讨以益气养阴活血法组方的"益肾活血方"对糖尿病肾病(diabetic nephropathy,DN)模型大鼠的作用机制。方法:用中药煎液灌服DN模型大鼠,生化检测实验大鼠24 h尿蛋白、血肌酐(Scr)、尿素氮(BUN)、血浆白蛋白(ALB)、血脂、血糖;Leica Qwin Plus图像分析系统测量肾小球直径;PAS染色观察大鼠肾脏病理;免疫组织化学检测肾组织TGF-β1表达。结果:该方能够降低实验大鼠24 h尿蛋白、Scr、BUN、升高ALB,减少肾小球直径及肾重,改善肾脏病理、降低肾组织TGF-β1表达,治疗组无论高、低剂量组与模型组相比,均具有统计学意义(P<0.05或P<0.01),但以高剂量组明显(P<0.01)。结论:益肾活血方可能通过影响DN大鼠肾组织中TGF-β1的表达,而改善上述生化指标,减轻肾脏病理损害,且与剂量相关。 Objective:To study the the mechanism of action of yiqi huoxue method in formula "" benefit of yishenhuoxue recipe" diabetic nephropathy (DN) model of of rats. Methods:Using traditional Chinese medicine Fried liquid filling DN model rats, serve biochemical tests 24 h urine protein experimental rats, creatinine (Scr), urea nitrogen (BUN), plasma albumin (ALB), blood fat and blood sugar; Leica Qwin Plus image analysis system;glomerular diameter measuring A PAS stain observation rat kidney pathological;Immunohistochemical (IHC) detection kidney tissues TGF-beta 1 expression. Results:The party can reduce experimental rats 24 h urine protein, Scr, BUN, higher ALB, reduce renal glomerulus diameter and heavy, improve renal pathology, reduce the kidney tissues TGF-beta 1 express,the treatment group whether high and low dose group, compared with model groups was statistically significant( P 〈 0.05 or P 〈 0. 01 ) ,but in the highdosage groups obvious( P 〈 O. 01 ). Conclusion: The benefit of yishenhuoxue recipe may through the effect of rats in the kidney tissues DN TGF-beta 1 expression, and improve the biochemical indicators, reduce kidney pathological damage dose-related.
