染料介导光氧化处理去细胞牛颈静脉带瓣管道的细胞毒性研究 被引量:1

摘要 目的:评价染料亚甲基蓝介导的光氧化处理去细胞牛颈静脉带瓣管道的细胞毒性。方法:(1)间接接触毒性试验:制备染料介导光氧化处理去细胞牛颈静脉带瓣管道浸提液,培养I.929细胞,然后采用MTT比色法测定相对增殖率。(2)直接接触毒性试验:将L929细胞与染料介导光氧化处理去细胞牛颈静脉带瓣管道直接接触培养,在不同时间观察细胞的生长状态。将L929细胞种植于染料介导光氧化处理去细胞牛颈静脉带瓣管道表面,用扫描电镜观察其生长情况。结果:染料亚甲基蓝介导光氧化处理去细胞牛颈静脉带瓣管道的性质稳定,细胞毒性程度为0~1级。L929细胞与瓣膜材料直接接触培养生长良好,形态学无明显改变。结论:染料介导光氧化处理去细胞牛颈静脉带瓣管道的细胞相容性良好。 Objective:To evaluate the cytotoxicity of acellular bovine jugular vein conduit treated with dye mediated photo oxidation. Methods:L929 cells were cultured in the extraction obtainted from acellular bovine jugular vein conduit treated with dye mediated photo oxidation to measure the relative growth rate by methyl-thiazolyl tetrazolium (MTT)assay; 1.929 cells were cultured directly with acellular bovine jugular vein conduit treated with dye mediated photo oxidation. The cell morphology was observed,and the cell number was counted;And the morphology of 1.929 cells cultured on the surface of acellular bovine jugular vein conduit treated with dye mediated photo oxidation was studied by scanning electron microscopy (SEM). Results:The property of acellular bovine jugular vein conduit treated with dye mediated photo oxidation was stable and the toxic levels ranged from 0 to 1 grade. There were no signs of cytotoxicity induced by acellular bovine jugular vein conduit treated with dye mediated photo oxidation,and the cell morphology was normal. Conclusion: Aeellular bovine jugular vein conduit treated with dye mediated photo oxidation has good eytocompatibility in vitro.
