摘要
在6种不同盐度(34、32、30、28、26和22)激活液、3种不同K+浓度(25 mmol/L、30 mmol/L和35 mmol/L)稀释液和不同保存时间(0 h、24 h、48 h、72 h和96 h)条件下,对日本鳗鲡(Anguilla japonica)精子的活力进行观察和测定。结果表明,激活液盐度为30时精子活率和运动精子百分比均最高,对应平均曲线运动速度(VCL)、平均直线运动速度(VSL)和平均路径速度均最高,由此推测自然界中日本鳗鲡产卵环境的盐度在30左右;稀释液中K+浓度为30 mmol/L时精子活率和运动精子百分比均最高,对应平均曲线运动速度(VCL)、平均直线运动速度(VSL)和平均路径速度(VAP)均显著高于25 mmol/L和30 mmol/L(P<0.05),据此推算出精子稀释液的最佳渗透压为796.68 kPa;随低温保存时间的延长,精子活率逐渐降低,稀释液中K+浓度为30 mmol/L时保存时间可达到96 h,并且精子活率与K+浓度为25 mmol/L、35 mmol/L时相比较,显著升高(P<0.05)。
Sperm motility of Anguillajaponica treated with six activation mediums with different salinities(34, 32, 30, 28, 26 and 22), three extender solution with different potassium ion concentration(25 mmol/L,30 mmol/L,35 retool/L) and different conservation time(0h, 24h, 48h, 72h and 96h) was observed and measured. The results showed that: when activation medium salinity was 30, both sperm motility and percentage of motile sperms were the highest, meanwhile the curvilinear velocity(VCL), the straight line velocity(VSL) and the angular path velocity(VAP) were the highest. It could be concluded that the spawning salinity of Anguillajaponica may be 30 in natural environment. When potassium ion concentration in extender solution was 30mmol/L, both sperm motility and percentage of motile sperms were the highest, meanwhile the curvilinear velocity(VCL), the straight line velocity(VSL) and the angular path velocity(VAP) were markedly higher than those under other concentration levels(P〈0.05). So the best osmotic pressure of extender solution may be 796.68kPa. As the conservation time extended, sperm motility reduced gradually. When potassium ion concentration in extender solution was 30 mmol/L, conservation time of sperm could reach 96 h and sperm survival rate was higher than in 25 mmol/L and 35 mmol/L potassium ion.
