摘要
目的: 探讨 bcl2 基因对人角膜上皮细胞生存过程的影响。方法: 用反转录聚合酶链式反应检测bcl2 m R N A 在人角膜上皮细胞中的表达情况。用荧光显微镜、电镜、流式细胞仪观察 bcl2 单克隆抗体对细胞凋亡发生的影响。结果: 用反转录聚合酶链式反应获得 332bp 的扩增带。荧光显微镜观察, 十个随机视野, 实验组与对照组凋亡细胞比例分别为 312% 和179% (u 检验, P< 001)。流式细胞仪检测的结果接近光镜下的结果, 凋亡细胞比例分别为 324% 和 198% 。电镜下可见典型的凋亡细胞。结论:人角膜上皮细胞有 bcl2 m R N A 的表达, bcl2 基因产物具有阻止人角膜上皮细胞凋亡发生的作用。
Purpose:To assess the role of bcl 2 in corneal epithelial cellular apoptosis.Methods:Reverse transcription polymerase chain reaction was used to determine bcl 2mRNA expression in human corneal epithelial cells.The effect of bcl 2 monoclonal antibody on human corneal epithelial cells was studied using light microscopy,electron microscopy and flow cytometry.Results:Bcl 2 mRNA was expressed in human corneal epithelial cells.bcl 2 antibody induced primary cultures of human corneal epithelial cells death with morphologic changes and DNA fragmentation consistent with apoptosis.Conclusions:Bcl 2 mRNA is expressed in corneal epithelial cells.Bcl 2 protein prevent human corneal epithelial cells from apoptosis.
