摘要
采用放射免疫法测定 8周龄、2 5周龄自发性高血压大鼠 (SHR) ,和正常血压Wistar -ky oto(WKY)大鼠血浆血栓素B2 (TXB2 )、6 -酮 -前列腺素F1α(6 -Keto -PGF1α)、前列腺素E2(PGE2 )、血管紧张素Ⅱ (AT -∏ )和肾髓质PGE2 浓度 ,并计算 6 -Keto/TXB2 比值。结果表明 :(1)各周龄SHR组血浆TXB2 浓度均显著高于WKY组 (P <0 0 0 1) ,且有随周龄增长而增高的趋势 ;(2 ) 2 5周龄SHR组血浆 6 -Keto -PGF1α浓度显著高于 8周龄SHR组和WKY组 (P <0 0 0 1) ,而 8周龄组与WKY组相比无显著差异 ;(3)不同周龄SHR组之 6 -Keto/TXB2 比值均明显低于WKY组 (P <0 0 0 1) ;(4 )血浆PGE2 和肾髓质PGE2 在SHR和WKY间差异均无显著性 ;(5)SHR血浆AT -Ⅱ浓度显著高于WKY(P <0 0 0 1) ;(6 ) 6 -Keto -PGF1α、TXB2 、PGE2 和AT -Ⅱ间未见明显的相关性。结果提示 :SHR血浆TXB2 、AT -Ⅱ增高 ,6 -Keto -PGF1α减少及 6 -Keto - /TXB2 比值降低 ,对高血压的发展起到一定的作用 ,而血浆和肾髓质中的PGE2 含量均无明显变化 ,揭示PGE2 并未发挥其抗高血压作用。
To study the relationship between the development of hypertension and prostagiands and angiotensin Ⅱ (AT Ⅱ) in SHR,radioimmunoassay was used to determine the concentrations of AT-Ⅱ,6-Keto-PGF 1α ,TXB 2 and PGE 2 in arterial plasma and the content of PGE 2 in renal medulla at different ages of spontaneously hypertensive rats (SHR) and normotensive control rats (WKY).The ratios of 6-Keto/TXB 2 were computed. The results showed that plasma TXB 2 concentratioin in SHR was significantly higher than that in WKY(P<0 001);Concentration of 6-Keto-PGF 1α in 25-week-old SHR was significantly higher than those in WKY and 8-week-old SHR (P<0 001),but no difference was found in 6-Keto-PGF 1α concentration of 8-week-old SHR and WKY; the ratios of 6-Keto /TXB 2 in both 25-week-old and 8-week-old SHR were lower than that in WKY(P<0 001).No obvious change was found in PGE 2 concentration of plasma and renal meduiia in SHR and WKY.AT-Ⅱof plasma in SHR was significantly higher than that in WKY(P<0 001).No interrelation was found between AT-Ⅱ,6-Keto-PGF 1α ,TXB 2 and their ratios,PGE 2 in plasma and in renal medulla. These results suggest that AT-Ⅱ plays an important role in generation of hypertension. The increase of TXB 2 and the decrease of 6-Keto/TXB 2 ratios are involved in the development of hypertension in SHR.PGE 2 concents both in plasma and in renal medulla had no obvious chandges in SHR,which suggests that PGE 2 didn't take its anti-hypertension role.
