TNF-α对类风湿关节炎滑膜细胞中MAPK的作用 被引量:7

摘要 目的 观察 T N Fα作用于类风湿关节炎( R A) 滑膜细胞后滑膜细胞中 M A P K 的激活。方法 3 ~5 代体外培养的 R A 滑膜细胞加外源性 T N Fα刺激后,裂解细胞,收获蛋白,进行 Western blot 分析。结果  T N Fα刺激后滑膜细胞中有多种蛋白发生酪氨酸磷酸化,其中以 M A P K最明显, M A P K 的激活呈时间和剂量相关。结论  T N Fα作用于 R A 滑膜细胞伴有明显的 M A P K 激活, M A P K 可能是 T N Fα作用于 R A 滑膜细胞后信号传导中的重要介质。 Objective To determine whether TNF α activates MAPK in rheumatoid synovial fibroblasts.Methods Cultured rheumatoid synovial fibroblasts were stimulated with exogenous TNF α,and then Western blot analysis of cell lysates was made.Results Multiple protein kinases were phosphorylated upon TNF α treatment.Induction of MAPK phosphorylation was time and dose dependent.Conclusion MAPK is likely to represent an important component in the cascade of signals that link TNF receptors to various TNF α elicited cellular responses in rheumatoid synovial fibroblasts.
