Factors Influencing Enzymatic Hydrolysis of Chrome Shavings from Leather Processing

摘要 选用1398蛋白酶对铬鞣革屑进行水解,从酶促反应动力学角度出发,研究了酶浓度、底物浓度、温度因素对酶解反应的影响,并通过正交试验,考察了各酶解条件的综合影响。结果表明:酶浓度和底物浓度超过一定范围时,表现为混合级反应,酶促反应受到抑制;反应温度在蛋白酶最适温度范围内时,酶活是影响反应速率的主要因素,反应温度超出最适范围很高时,温度成为影响反应速率的主要因素;1398蛋白酶最佳酶解条件为:底物浓度2%,反应温度60℃,酶浓度2%,氮收率达到60%左右。 The chrome shavings were used as the raw material and was enzymatic hydrolyzed by the neutral protease 1398. The main factors effecting the hydrolysis reaction were investigated, including dosage of enzyme, hydrolysis temperature, solid/liquid ratio from perspective of kinetics of enzyme - catalyzed reaction, and the combined effect of the enzyme hydrolysis factors were researched by orthogonal test. The results show that enzymatic reaction is restrained and performed as mixed - level reaction with dosage of enzyme and solid/liquid ratio over a certain range. Enzyme activity is the main factor with the hydrolysis temperature during the protease' s optimum temperature range, and hydrolysis temperature is the determinant when it beyond the range highly. The optimum enzymatic conditions are as foUows: solid/liquid ratio is 2%, hydrolysis temperature is 60℃, enzyme dosage is 2%, and the nitrogen recovery is about 60%.
