血管外膜炎症大鼠核转录因子NF-κB、白介素-6、白介素-10的表达及通心络的干预研究 被引量:6

摘要 目的:探讨血管外膜炎症在动脉粥样硬化(atherosclerosis,AS)形成过程中的作用及通心络的保护作用。方法:将20只健康Wistar Kyoto大鼠采用套管法建立大鼠左颈动脉外膜损伤炎症模型后随机分为两组,分别为外膜组和通心络组,每组各10只。分别予以生理盐水和通心络超微粉剂(TXL)400 mg/(kg.d)灌胃,共2周。2周后分离血浆检测细胞因子白介素-6(IL-6)、白介素-10(IL-10)水平。包管段切片通过免疫组化法测定核转录因子NF-κB的表达。结果:2周后,外膜组IL-6(147.40±4.65)mg/L、IL-10(44.78±1.27)mg/L;通心络组血清IL-6(143.85±2.66)mg/L、IL-10(42.35±2.31)mg/L,两者含量均明显低于外膜组(P<0.01);NF-κB表达亦明显下降(P<0.005)。结论:通心络能减轻炎症病变,具有血管保护作用,这一作用可能部分通过抑制核转录因子NF-κB的活化来实现。 Objective: To explore adventitial inflammation in atherosclerosis(atherosclerosis,AS) the important role during the formation and the intervention of Tongxinluo.Methods: Twenty healthy male Wistar rats(12-weeks-old) were establishment of rat left common carotid artery(LCA) Inflammatory model,randomly divided into 2 groups,model group and Tongxinluo treat group.Tongxinluo(TXL) group were poured Tongxinluo 400 mg/(kg·d);model group were poured saline 400 mg/(kg·d) in stomach.At two weeks treatment,the levels of IL-6,IL-10 in plasma were measured by ELISA;The levels of NF-B were stained by immunohistochemical method.Results: After 2 weeks treatment,the levels of IL-6(147.40±4.65),IL-10(44.78±1.27) in the outer membrane.Compared with the Tongxinluo group displayed a significant reduction in the levels of IL-6(143.85±2.66),IL-10(42.35±2.31)(P0.01) and NF-κB(P0.005).Conclusion: Tongxinluo with a vascular protective effect can reduce atherosclerotic lesions which possibly through Inhibition of nuclear transcription factor NF-B p65 activation.
