高效液相色谱法测定珍菊降压片中盐酸可乐定的含量

Determination of clonidine hydrochloride in Zhenju Jiangya Tablets by HPLC

目的:建立以高效液相色谱法(HPLC)测定珍菊降压片中盐酸可乐定含量的方法。方法:采用Diamonsil C18(4.6 mm×250 mm,5μm)色谱柱,流动相为0.22%辛烷磺酸钠-甲醇-磷酸(560∶440∶1,用1 mo/L氢氧化钠溶液调节pH至3.0),检测波长为220 nm,流速为1.0 ml/min,进样量为20μl,柱温为室温。结果:盐酸可乐定在1.0～10.0μg/ml范围内成良好的线性关系(r=0.999 9,n=6),平均回收率为99.4%,RSD为0.93%。结论:本方法精密、准确,可用于珍菊降压片中盐酸可乐定的质量控制。

Objective： To develop the HPLC method for the determination of clonidine hydrochloride in Zhenju Jiangya Tablets.Methods： The method was performed on Diamonsil C18（4.6 mm×250 mm,5 μm） column.The mobile phase consisted of 0.22% Sodium 1-octanesulfonate-methanol-phosphoric acid（560∶440∶1,adjusted to pH 3.0 by 1 mol/L sodium hydroxide solution） at the UV detection wavelength was 220 nm,the flow rate of 1.0 ml/min,the injection volume was 20 μl and the column temperature was room temperature.Results： The linear range of clonidine hydrochloride was 1.0-10.0 μg/ml（r=0.999 9,n=6）,with average recovery of 99.4%（RSD=0.93%）.Conclution： The established method is accurate and suitable for the quality control of Zhenju Jiangya Tablets.
