湖州市甲型H1N1流感病毒株的HA基因分析

摘要 目的:研究2010年湖州市甲型H1N1流感分离株(A/Huzhou/2010(H1N1))的HA基因特征。方法:采用MD-CK细胞对患者咽拭子和漱口液标本进行病毒分离,逆转录-聚合酶链反应(RT-PCR)扩增病毒HA基因并进行序列测定,用DNAstar和MegAlign等软件分析处理。结果:湖州市2010年甲型H1N1分离株的HA基因为1739个核苷酸,与湖州市2009年的分离株及WHO2010/2011推荐的甲型H1N1疫苗株相比,没有核苷酸的插入与丢失,氨基酸同源性分别为99.0%和98.9%。进化树分析显示,本次分离株与湖州市2009年的分离株及WHO2010/2011推荐的疫苗株非常相似。结论:甲型H1N1流感分离株的分子生物学特征及变异特征分析,对制定科学有效的防控策略具有重要意义。 Objective:To study the genetic characteristc of HA gene of Influenza A(H1N1)from Huzhou city in 2010.Methods: Respiratory specimens were collected and the influenza viruses were isolated using the MDCK cell.Nucleic acids were extracted from the isolated viruses and the HA gene were amplified by reverse transcription polymerase chain reaction(RT-PCR).The amplicons were sequenced and the data were subjected to the alignment analyses using the softwares of DNAstar and MegAlign.Results: HA regions of the isolate were 1739 bp.When compared with the viruses of A/HuZhou/09(H1)and A/California/07/2009(H1N1),there was no deletion or insertion of nucleotide acid.And the homology of Amino acids were 99.0% and 98.9% respectively.Phylogenetic analysis revealed that the evolutional relationship of the HA gene of A/HuZhou/2010(H1N1)between A/HuZhou/09(H1)and A/California/07/2009(H1N1) were very near.Conclusion: The variation and molecular biology analyses of the influenza A(H1N1) virus are of great imporance to the development of scientific and effective prevention and control strategies of influenza.
