摘要
目的:探讨自由基清除剂依达拉奉对坐骨神经损伤后神经功能及脊髓脂质过氧化反应的影响。方法:Wistar大鼠48只,随机分为3组:坐骨神经挤压伤组、依达拉奉治疗组、假手术组。分别于7、14、21、28 d检测各组大鼠坐骨神经功能指数(SFI)、脊髓内过氧化物歧化酶(SOD)和丙二醛(MDA)的变化。结果:伤后各组大鼠SFI均降低,挤压伤组大鼠SFI较依达拉奉治疗组低(P<0.05),神经功能恢复较治疗组缓慢。伤后挤压伤组大鼠脊髓内SOD活性升高,依达拉奉治疗组大鼠脊髓内SOD活性与假手术组相比升高不明显(P>0.05)。伤后挤压伤组大鼠脊髓内MDA含量上升明显,依达拉奉治疗组大鼠脊髓内MDA含量在各个时间点均显著低于挤压伤组大鼠脊髓内MDA含量(P<0.05)。结论:依达拉奉能通过抑制坐骨神经损伤后引发的脊髓脂质过氧化反应对神经元发挥保护作用,对坐骨神经功能恢复有促进作用。
Objective:To investigate the relationship between edaravone and lipid peroxidation after sciatic nerve injury.Methods:48 Wistar rats were divided into 3 group randomly:sciatic nerve crush injury group,edaravone-treated group and sham operation group.To detect sciatic function index(SFI),superoxide dismutase(SOD) and malonydialdehyde(MDA) at 7 d,14 d,21 d,28 d after operation.To observe the effects of edaravone on lipid peroxidation after sciatic nerve injury.Results:SFI was decreased in all the groups after the operation,SFI in the crush injury group was lower than that in the edaravone-treated group(P0.05).The sciatic nerve function recovered slowly in the crush injury group.The activity of SOD was increased in the crush injury group.There was no difference between the edaravone-treated group and the sham operation group(P0.05).The MDA content in the spinal cord was increased obviously in the crush injury group.The MDA content in spinal cord of rats in the edaravone-treated group was significantly lower than that in the crush injury group at different time points(P0.05).Conclusion:Edaravone can inhibit lipid peroxidation after sciatic nerve crush injury in rats to protect neurons,so that which can enhance the nerve functional recovery.
