缺氧条件下绒毛外滋养细胞中Cyr61和NOV mRNA表达研究

摘要 目的检测CoC l2化学缺氧条件下,绒毛外滋养细胞中促血管生成蛋白Cyr61和NOV的转录水平表达情况,探讨其在子痫前期发病机制的研究。方法采用实时荧光定量逆转录聚合酶链反应(qRT-PCR)技术检测绒毛外滋养细胞中Cyr61和NOV mRNA的表达。结果 (1)促血管生成蛋白Cyr61和NOV在绒毛外滋养细胞中表达。(2)与正常对照组相比,缺氧条件下Cyr61和NOV转录水平降低(P<0.05)。结论 Cyr61和NOV的表达下调可能与胎盘血管生成障碍有关,从而参与了子痫前期的发病有关。 Objective:To detected expression levels of the proangiogenic protein Cyr61 and NOV in extravillous trophoblasts under hypoxia,using cobalt chloride(CoCl2) as hypoxic mimetic.Methods: Quantitative polymerase chain reaction(qRT-PCR) was implied to investigate expression of Cyr61 and NOV in extravillous trophoblasts.Results:(1) The proangiogenic protein Cyr61 and NOV were founded in extravillous trophoblasts.(2)Compared with control,expression levels of Cyr61 and Nov mRNA in trophoblast cells cultured under hypoxic condition(P0.05).Conclusion: Down-regulation of Cyr61 and NOV might result in failure of placental vascular formation and play role in the pathogenesis of preeclampsia.
