摘要
通过超声、离心、透析的方法将富硒花生中的有机硒和无机硒分离开,有机态硒以蛋白质和多糖为主要分离对象,采用火焰原子吸收光谱法对硒水平进行测定。结果表明:富硒花生中硒含量为0.165 3μg/g,蛋白提取率为23.39%,多糖提取率为11.32%,硒蛋白是硒的主要赋存形态,其结合的硒量达到富硒花生有机硒总量的73.39%,总硒量的59.89%。抗氧化性分析结果表明,粗多糖相对于纯化多糖和蛋白提取物,具有更长的诱导时间、更高的DPPH自由基清除能力和更高的还原力。
The organic selenium and inorganic selenium of selenium enriched peanuts were separated with ultrasonic, centrifuge and dialysis method. The main target for organic selenium is protein and polysaccharide. The content of selenium of the sample was determined by flame atomic absorption spectrometry. The result shows that the extraction of protein was 11.32% and the extraction of polysaccharide was 11.32%. The content of selenium in selenium enriched peanuts was 0. 1653 μg/g. Selenoprotein was the major format of selenium in the peanuts, it combined up to73.39% of organic selenium, and 59.89% of the total selenium in the peanuts. The result of antioxidant analysis showed that the crude polysaecharide had longer induction time, higher DPPH free - radical scavenging activity and reducing activity than purified polysaccharide and protein extracts.
