复方丹参注射液对门脉高压性胃病模型大鼠胃黏膜上皮细胞凋亡的影响

Effects of the Compound Dansen Injection on apoptosis of gastric mucosa in rats with portal hypertension gastropathy

目的 观察复方丹参注射液对门脉高压性胃病（PHG）模型大鼠胃黏膜上皮细胞凋亡的影响.方法 采用一期部分门静脉缩窄法制备PHG大鼠模型,雄性SD大鼠70只,电脑随机抽取分为3组：A组（PHG造模＋复方丹参注射液）30只、B组（PHG造模＋0.09%氯化钠溶液）30只、C组（假手术＋0.09%氯化钠溶液）10只.造模后各时段（5、10、15、20、30、45d）处死大鼠,取胃底黏膜组织,检测各组大鼠胃黏膜组织病理学变化、上皮细胞凋亡基因Bcl-2、Bax mRNA表达及原位末端标记（TUNEL）染色测凋亡指数（apoptotic index,AI）的变化.结果 A组的胃黏膜破坏明显减轻.B组胃黏膜组织中Bcl-2表达水平则显著下调,在第20天达最低值0.12±0.09,其后趋于稳定;Bax表达水平显著上调,并在第20天达到高峰6.12±0.39,其后趋于稳定;AI值上升,第20天最高（11.08±1.62）%,后趋于稳定.A组小鼠Bcl-2及Bax表达水平及AI值发生改变,20d时各值分别为：（1.09±0.21）、（1.12±0.17）、（5.32±1.07）%,各时间点与B组比较差异均有统计学意义（均P＜0.01）.而C组20d时Bcl-2、Bax表达水平、AI值分别为：（0.98±0.10）、（0.86±0.11）、（4.32±0.08）%,与A组相比差异无统计学意义（P ＞0.05）,但与B组比较差异有统计学意义（均P＜0.01）.结论 复方丹参注射液能明显减轻PHG模型大鼠胃黏膜损伤并抑制上皮细胞凋亡,对PHG模型大鼠胃黏膜具有保护作用.

Objective To investigate the effects of the Compound Dansen Injection on apoptosis of the gastric mucosa in rats with portal hypertension gastropathy. Methods The portal hypertension gastropathy was induced by partial portal vein ligation in rats. Seventy male Sprague Dawley rats were randomly divided into 3 groups： in group A 30 rats with portal hypertension gastropathy were treated with Compound Dansen Injection; in group B 30 rats with portal hypertension gastropathy were not treated; 10 normal rats served as controls （group C）. The animals were sacrificed at d5, d10, d15, d20, d30 and d45, and gastric mucosa samples were taken. The hisfopathological changes of gastric mucosa were observed. The mRNA expression level of Bcl-2 and Bax were detected by reverse transcription polymerase chain reaction （RT-PCR） and the apoptotic indexes （AI） were measured by TUNEL. Results The lower degree of mucosal damage was observed in group A. In B group the BCL-2 expression level decreased to （0.12 ± 0.09）at d20; the BAX expression level increased to （6.12 ±0.39）at d20. The apoptotic indexes increased to （ 11.08 ±1.62）% at d20. Compared to group B the expression of BCL-2, BAX mRNA and AI in group A markedly changed [（1.09 ±0.21 ）%, （1.12 ±0.17）% and （5.32 ±1.07）%, respectively] at d20 （ P〈0.01 ）; there were no differences of above parameters between groups A and group C [（0.98 ±0.10）%,（ 0.86 ±0.11）% and （4.32 ±0.08）%, respectively]at d20 （P 〉0.05 ）. there were significantly differences between group C and group B （P〈0.01）. Conclusion The Compound Danshen Injection has protective effects on the gastric mucosa in rats with portal hypertension gastropathy.
