ω-3多不饱和脂肪酸对胃癌细胞系生长抑制作用及其机制的研究 被引量:8

摘要 目的:观察ω-3多不饱和脂肪酸(ω-3PUFAs)对MGC803低分化胃癌细胞系凋亡的影响,并对其机制进行探讨。方法:采用四甲基偶氮唑蓝(MTT)观察细胞增殖率、DNA电泳和流式细胞术(FCM)检测细胞凋亡情况、蛋白质印迹法检测凋亡蛋白PARP-1和AIF的表达。结果:二十碳五烯酸(EPA)和二十二碳六烯酸(DHA)通过诱导细胞凋亡显著抑制胃癌细胞的生长,呈时间和剂量依赖关系。DHA促进细胞凋亡的作用强于EPA。40μg/mL EPA和DHA作用MGC803细胞24 h后,凋亡相关蛋白PARP-1和AIF表达均较对照组显著增加。结论:ω-3 PUFAs通过诱导细胞凋亡抑制胃癌细胞的增殖,其机制可能通过PARP-1/AIF凋亡通路实现。 OBJECTIVE: To investigate the effect of ω-3 polyunsaturated fatty acids (ω-3 PUFAs) on the apoptosis of hu man gastric cancer cell line MGC803, and study the potential mechanisms. METHODS: Cell growth and apoptosis were analyzed with MTT assay, DNA electrophoresis and flow cytometry. Western blot was used to analyzed PARP-1 and AIF. RESULTS: Both EPA and DHA markedly inhibited the MGC803 cell growth and induced apoptosis in a time and concentration de pendent manner. The effect of DHA on apoptosis of cells was stronger than that of EPA. After incubation of the cells with 40 μg/mL EPA or DHA for 24 hours, the level of PARP-1 and AIF significantly increased. CONCLUSIONS: ω-3 PUFAs inhibit the growth of gastric cancer cells through promoting apoptosis. It in dicate that EPA/DHA appear to be mediated apoptosis of MGC803 cells line in part by the activation of PARP 1/AIF pathways.
