重组人骨保护素(rhOPG-Fc)联合降钙素治疗家兔激素性股骨头坏死的实验研究

摘要 目的:研究重组人骨保护素(rhOPG-Fc)联合降钙素对兔激素性股骨头坏死的治疗作用。方法:进行人类OPG-Fc融合蛋白毕赤酵母表达株的构建和鉴定。24只实验白兔被随机分为3组。治疗组:激素注射造模成功后,采用rhOPG-Fc联合降钙素治疗;激素组:激素注射造模;对照组:注射生理盐水。治疗结束后对3组兔股骨头进行组织学检查和OPG/OPGL免疫组织化学染色。结果:(1)经鉴定证实重组人骨保护素(rhOPG-Fc)构建成功;(2)与激素组比较,治疗组毛细血管数、管径及骨陷窝空缺百分比均有显著改善(P<0.05);(3)与激素组比较,治疗组OPG的表达水平明显升高(P<0.05),而OPGL的表达水平明显下降(P<0.05),OPG/OPGL的比值明显升高(P<0.05)。结论:通过对OPG/OPGL的调节,重组人骨保护素(rhOPG-Fc)联合降钙素可有效治疗兔激素性股骨头坏死。 Objective:To study the effect of recombinant human osteoprotegerin(rhOPG-Fc) combined with calcitonin on treatment of the femoral head necrosis in rabbits induced by hormone.Methods:Recombinant human osteoprotegerin secretory expression in P.pastoris was constructed and identified.Twenty-four experimental rabbits were randomly divided into 3 groups.Treatment group(6 rabbits): rhOPG-Fc combined with calcitonin was use to treat after model was constructed by hormone;hormone group(6 rabbits): model was constructed by hormone;control group: saline was injected.The femoral heads and expression of OPG/OPGL in 3 groups were examination through histological and immunohistochemistry.Results:(1) The recombinant human osteoprotegerin(rhOPG-Fc) was successfully constructed through identified;(2) Compared with the hormone group,the number and diameter of capillaries,percentage of vacant lacunae in treatment goup improved significantly(P0.05);(3) Compared with the hormone group,the expression of OPG increased significantly(P0.05),and the expression of OPGL decreased significantly(P0.05).The ratio of OPG/OPGL increased significantly(P0.05).Conclusion:Through the regulation of OPG/OPGL,recombinant human OPG(rhOPG-Fc) combined with calcitonin get a good effective on treatment of the femoral head necrosis in rabbits induced by hormone.
