Effects of isatin on substantia nigra BAX gene expression in MPTP-induced PD model in mice

目的探讨2,3吲哚醌（isatin,ISA）对MPTP制备的C57BL/6J小鼠帕金森病（PD）模型黑质中BAX基因表达影响。方法 C57BL/6J小鼠随机分为五组,空白对照组,MPTP损伤组,ISA低（50 mg/kg,ig）、中（100 mg/kg,ig）、高剂量（200 mg/kg,ig）预处理＋MPTP组。结果与对照组相比,MPTP损伤的小鼠黑质中BAX免疫反应阳性表达增加（P〈0.01）;在ISA 100 mg/kg和200 mg/kg预处理组与MPTP损伤组相比,黑质中BAX免疫反应阳性表达显著降低（P〈0.05）。结论表明ISA对MPTP造成的PD小鼠有明显的保护作用。

Objective：To explore the effect of isatin on substantia nigra（SN） BAX gene expression in MPTP-induced PD model in mice.Methods：C57BL/6J mice were divided into 5 groups：① control group;② MPTP group;③ ISA 50 mg/kg ig ＋ MPTP group;④ ISA 100 mg/kg ig ＋ MPTP group;and ⑤ ISA 200 mg/kg ig ＋ MPTP group.Then immunohistochemical techniques was used to observe the BAX gene＇s expression in substantia nigra in each mouse.Results：The results showed that compared with the control group,the BAX immunoreactive expression in SN was higher in MPTP injury mice（P0.01）;in ISA 100 mg / kg and 200 mg / kg pretreatment group,the BAX immunoreactive expression in SN was significantly reduced compared with the MPTP injury group（P0.05）.Conclusion：It is shown that the ISA has the protective effect on MPTP-induced PD model in mice.
