清肝调脂饮对非酒精性脂肪性肝炎大鼠胰岛素抵抗的影响 被引量:8

摘要 目的探讨清肝调脂饮对非酒精性脂肪性肝炎(NASH)大鼠胰岛素抵抗的干预机制。方法建立高脂饲料诱导的Wistar大鼠NASH模型并随机分组,清肝调脂饮低、中、高剂量组和西利宾胺组分别灌胃相应剂量药物治疗。实验12周后,检测各组大鼠血清TG、TC、葡萄糖、瘦素、FFA、INS;肝组织行脂肪染色、糖原染色并作定量分析。结果与模型组比较,各用药组均能不同程度改善以上各指标(P<0.05,P<0.01),其中,在降低TG、葡萄糖、INS、FFA方面,中药高剂量组均明显优于西药对照组(P<0.05)。结论清肝调脂饮促进游离脂肪酸代谢与肝内TG分解;改善胰岛素抵抗(IR)及瘦素抵抗(LR)。与西利宾胺比较,清肝调脂饮在抑制NASH的IR方面具有明显的疗效优势。 Objective To observe the intervention mechanism of Qinggantiaozhi decoction on non-alcoholic steatohepatitis insulin resistance(IR) by animal experiment.Methods The model of NASH was established by feeding Wistar rats with high fat diet and were randomly divided into 4 groups.The groups of Qinggantiaozhi decoction in low dose,middle dose and high dose,and the control group of silybin,were treated by corresponding medicine.After 12 weeks,all the rats were put to death.The serum TG,TC,GLu,Leptin,FFA,INS were detected.The liver tissue was stained by both fat and glycogen.All the above positive results were analyzed by quantitative analysis.Results After analyzing the experimental data,all the above indexes are improved in the treated groups compare with those of in model group(P0.05,P0.01).The index of TG,GLu,FFA,INS is reduced in high dose group of Qinggantiaozhi decoction than that in silybin group(P0.05).Conclusion There maybe several mechanisms for Qinggantiaozhi decoction to prevent and cure NASH.The most important one is to correct disordered blood-fat and reduce the deposition of fat in hepatic cell through improving the resistance of insulin and leptin,promoting metabolism of FFA and TG in hepatic cell.It demonstrates that the Qinggantiaozhi decoction can obviously reduce the blood-fat and improve the iconography appearance of the nonalcoholic steatohepatitis.
