摘要
目的探讨肿瘤坏死因子受体相关因子-5(tumor necrosis factor receptor-associated factor-5,TRAF-5)在炎症性肠病患者外周血单个核细胞和血浆的表达,比较炎症性肠病患者TRAF-5表达和正常对照者的差异,分析血浆TRAF-5表达与内镜下疾病活动性的相关性。方法应用酶联免疫吸附试验(enzyme-linked immuno sorbent assay,ELISA)分析炎症性肠病患者和正常对照者血浆中TRAF-5蛋白的表达,SYBR-green real time PCR方法分析炎症性肠病患者和正常对照者外周单个核细胞中TRAF-5 mRNA的表达。数据处理使用GraphPad Prism 5。结果克罗恩病患者(P=0.003)和溃疡性结肠炎患者(P=0.000)血浆TRAF-5水平显著高于正常对照者,但是克罗恩病患者(r=-0.167,P=0.194)以及溃疡性结肠炎患者(r=0.008,P=0.952)血浆TRAF-5表达水平和内镜下疾病活动指数均无显著的相关性。克罗恩病患者(P=0.000)和溃疡性结肠炎(P=0.000)患者外周血单个核细胞TRAF-5 mRNA表达均显著高于正常对照者。结论炎症性肠病患者血浆和外周血单个核细胞中均存在TRAF-5的激活,但血浆TRAF-5的高表达不能反映内镜下疾病的活动程度。
Objective To investigate the expression of tumor necrosis factor receptor-associated factor-5(TRAF-5) in peripheral blood mononuclear cells and plasma in patients with inflammatory bowel disease,to find the difference between the expression of TRAF-5 in patients with inflammatory bowel disease and in healthy controls,and to analyze the correlation between plasma TRAF-5 levels and endoscopic disease activity.Methods Enzyme-linked immuno sorbent assay(ELISA) was applied to evaluate plasma TRAF-5 levels in patients with inflammatory bowel disease and in healthy contols.SYBR-green real time PCR was applied to access TRAF-5 mRNA expression in peripheral blood mononuclear cells in patients with inflammatory bowel disease and healthy controls.Data were analyzed using Graph Pad Prism 5.Results Plasma levels of TRAF-5 were significantly higher in Crohn's disease(P =0.003) and ulcerative colitis(P =0.000) than in healthy controls.However,no sinificant correlation was indicated between plasma TRAF-5 and endoscopic disease activity in patients with Crohn's disease(r = -0.167,P =0.194) or in patients with ulcerative colitis(r =0.008,P =0.952).Furthermore, TRAF-5 mRNA expression in peripheral blood mononuclear cells was significantly higher in patients with Crohn' s disease and ulcerative colitis than in healthy controls(all P =0.000).Conclusion TRAF-5 activated in plasma and peripheral blood mononuclear cells in patients with inflammatory bowel disease.However,plasma TRAF-5 levels can not reflect endoscopic disease activity.
