摘要
目的 构建抗人血管生长素基因工程抗体文库。方法 采用PCR等分子生物学实验手段从小鼠杂交瘤细胞中获取抗人血管生长素抗体可变区重、轻链基因,通过噬菌体表面呈现技术表达抗体。结果 ①获取了抗人血管生长素抗体可变区基因片段(ScFv) ,②得到了抗人血管生长素基因工程抗体文库。结论 本研究建立了一种构建高效抗人血管生长素抗体文库的方法,该抗体库在肿瘤的临床治疗上具有广阔的应用前景。
Objective To construct library of the phage genetic engineering antibody of anti-human angiogenin. Methods By using molecular biological methods such as polymerase chain reaction, etc, both heavy and light chain genes of anti-human angiogenin antibody were prepared from mouse angiogenin hybridomas. Antibodies were expressed by using the technique of phage surface display libraries. Results ①The DNA fragments of variable region of anti-human angiogenin antibody were obtaines. ②The library of phage genetic engineering antibody of anti-human angiogenin were constructed. Conclusion Construction of the library of anti-human angiogenin antibody is established. This library of antibodies may provide an alternative way in the treatment of tumor.
