摘要
目的:观察苦参碱作用于肝癌HepG2细胞后,倒置显微镜和多功能显微镜下的细胞形态结构改变。方法:倒置显微镜下观察活细胞形态;多功能显微镜下观察瑞氏染色和苏木精-伊红(HE染色)染色的细胞形态。结果:未用药细胞具有肿瘤细胞的典型的形态特征,苦参碱处理细胞后,细胞数量减少,分散生长,形态呈多形性。细胞浆内充满圆形透明的颗粒。细胞在倒置显微镜下形态似上皮细胞样,细胞间相互连接,密集成片生长,细胞浆内未见透明颗粒。结论:苦参碱对HepG2细胞的生长增殖有抑制作用,是HepG2细胞的某些恶性增殖表型减弱或消失,说明苦参碱可能是一种很好的抗肿瘤药。
OBJECTIVE:To observe the morphological change of HepG2 cells under inverted microscope and multifunctional microscope after exposure to matrine.METHODS:The morphology of the viable HepG2 cells were observed under inverted microscope and that of the Wright stained and hematoxylineosin(HE staining) stained HepG2 cells were observed under multifunctional microscope.RESULTS:None-treated HepG2 cells assume typical morphological feature of tumor cells,however,matrine-treatment resulted in reduced number of HepG2 cells,and the cells showed a dispersed growth pattern and appeared pleomorphic,with spherical transparent particles filled within cytoplasm.Under inverted microscope,the HepG2 cells assumed the shape of epithelial cells,with cells interconnected and densely scattered into pieces yet without transparent particles within cytoplasm.CONCLUSION:Matrine has inhibitory action on the growth of HepG2 cells.The attenuation or disappearance of phenotypes of the malignant proliferation is suggestive of the possibility of matrine as an effective anticancer drug.
