摘要
目的进行肝癌Autotaxin(ATX)cDNA部分序列测定,观察ATXmRNA在肝癌组织中的表达程度,以探讨ATX在肝癌转移机理中的作用。方法提取肝癌7721细胞株、5例正常肝组织、32例肝癌组织RNA,参照黑色素瘤ATXcDNA序列设计引物,经逆转录聚台酶链反应(RT-PCR)扩增肝癌ATX,以卜actin基因为内参照进行ATXmRNA表达程度分析,同时7721细胞株RT-PCR产物进行cDNA序列测定。结果肝癌7721细胞株32例肝癌组织5例正常肝组织中均有AutotaxinmRNA表达,其cDNA序列测定(33hp)显示与黑色素瘤具有99.7%的同源性;癌组织ATXmRNA表达显著高于正常肝组织(分别为0.68±0.1和0.31±0.08)。结论肝癌及正常肝组织表达ATXmRNA,ATX可能参与了肝癌的浸润与转移过程,在其中扮演了重要角色。
Objective To know if ATX is expressed in hepatocellular carcinoma. Methods 7721hepatoma cell lines in culture, 5 normal liver tissues, and 32 histologically verified HCC specimens wereobtained. Semiquantitative reverse transcription polymerase chain reaction was used to detect mRNA expression of ATX. Results The cDNA sequence of RT--PCR product of 7721 hepatoma cell lines wasmeasured, it showed that nucleotides measured were 332hp. ATX cDNA nucleotide wnuence in HCC wasfound to share 99.7% homology with that in Melanoma. ATX was expressed in all of the 32 HCC and 5normal liver tissues. The mean expression level of ATX bine in HCC samPles was higher than that innormal liver tissues (0.68 ± 0.1 vs 0.3 ± 0.08, p<0.01). Conclusion ATX bine was found to be in existencein HCC and normal liver tissues. The homology of ATX cDNA sequence between HCC and humanMelanoma was 99.7%, and ATX may play an important role in HCC metastatic mechanism.
