脂质体-C-erbB2反义脱氧寡核苷酸对卵巢癌细胞的作用 被引量:8

Effect of Liposome C erbB2 Antisense Oligodeoxynucleotides on Human Ovarian Cancer Cells

摘要 目的 探讨脂质体 Cerb B2 反义脱氧寡核苷酸( 脂质体 Cerb B2 S O D Ns) 对卵巢癌细胞系 Cerb B2 癌基因表达及细胞增殖的影响。方法 应用流式细胞仪技术,3 H胸腺嘧啶核苷掺入方法,观察脂质体 Cerb B2 S O D Ns 对卵巢癌细胞系 S K O V3 Cerb B2 癌基因表达及细胞生长、细胞周期的作用。结果 脂质体 Cerb B2 S O D Ns 能够降低 Cerb B2 的表达,并抑制细胞生长;脂质体 Cerb B2 S O D Ns降低细胞 Cerb B2 癌基因表达的作用效率较单用 Cerb B2 反义脱氧寡核苷酸( Cerb B2 S O D Ns)增强约40 倍。结论  Cerb B2 癌基因的反义调控在卵巢癌基因治疗中有一定作用;脂质体可有效增强 Cerb B2 S O D Ns 调控细胞 Cerb B2 癌基因表达的作用。 Objective To explore the effects of liposome C erbB2 antisense phosphorothioate oligo deoxynucleotides(S ODNs)on C erbB2 protooncogene expression and cell proliferation in human ovarian cancer cells. Methods The effects on C erbB2 protooncogene expression, cell proliferation and cell cycle in human ovarian cancer cells were studied by flow cytometry and 3H thymidine incorporation. Results Liposome C erbB2 S ODNs could reduce C erbB2 expression and inhibit cell proliferation in human ovarian cancer cells; the effectiveness of liposome C erbB2 S ODNs on the expression of C erbB2 was much higher than that of C erbB2 S ODNs, about 40 times. Conclusions The data in this study suggest that antisense therapy is an useful method of gene therapy in ovarian cancer. The effectiveness of C erbB2 S ODNs could be greatly increased by C erbB2 S ODNs encapsulated in liposomes.
