转基因人肝癌细胞株(BEL 7404/TNF)的建立及其生物活性的初步研究 被引量:3

摘要 目的 :研究转基因肝癌细胞和其来源肝癌细胞株的生物学特性的差异。方法 :构建含人肿瘤坏死因子α基因 ( TNFα基因 )的重组逆转录病毒载体 ,将其导入人肝癌细胞株 BEL 74 0 4。通过 MTT比色法测定比较转基因肝癌细胞株及未转基因肝癌细胞株的生物活性。结果 :成功建立转基因肝癌细胞株。MTT比色法测定结果表明转基因的肝癌细胞株生长速度显著减慢。结论 :TNF基因转入肝癌细胞后 。 Objective:To study the difference of biological character between transfected TNF gene liver cancer cell line and original cell line Methods:Establishing recombinant retrovirus vector containing TNF gene Transfecting the vector into liver cancer cell line BEL7404 And then the biological character of transfected TNF gene cell line was compared with its original cell line by MTT assay Results:Transfected TNF gene cell line was established successfully Proliferative speed of transfected gene cell line was remarkable slower than that of the original cell line Conclusion:Malignant degree of liver cancer cell line was decreased after TNF gene was transfected into it
