摘要
目的探讨神经酰胺(Cer)对小鼠皮层神经元噻唑蓝(MTT)代谢率的影响。②方法以原代培养的26只小鼠大脑皮层神经元为模型,观察Cer不同浓度(0,50,100,200,500,1000,2000,5000μmol/L),不同的作用时间(4,8,12,16,24,36h)对其MTT代谢率的影响。③结果MTT代谢率随着Cer浓度的增加及作用时间的延长明显下降(t=2.08~2.14,P<0.05)。④结论Cer可损伤神经元线粒体功能,从而导致神经元死亡。
Objective\ To explore the effect of ceramide on MTT metabolism rate of mouse cortical neuron.\ Methods The effect of eramide at different concentrations (0,50,100,200,500,1 000,2 000,5 000μmol/L) and different acting times (4,8,12,16,24,36h) on the metabolism rate of MTT in the primary cell culture of 26 mice cortical neuron were studied.\ Results With the increase of ceramide concentration and acting time, the metabolism rate of MTT decreased (t=2.08-2.14,P<0.05). Conclusion\ Ceramide can damage the function of neuron mitochondrion and then lead to the death of neurons.
