摘要
目的 探讨上皮生长因子( E G F) 、碱性成纤维细胞生长因子(b F G F) 和苏拉明( Suramin) 对体外培养人脑膜瘤细胞生长的影响。方法 通过细胞计数、 M T T 法及流式细胞技术检测,对体外培养手术切除的10 例脑膜瘤组织进行 E G F、b F G F 及 Suramin 干预实验。结果 苏拉明对脑膜瘤细胞的生长有明显的抑制作用,使细胞停滞于 G1/ G2 及 S 期,并可阻止肽类生长因子对细胞的生长刺激作用。结论 苏拉明对体外培养脑膜瘤的生长抑制作用可能通过中和肽类生长因子及影响细胞周期分布实现。
Objective\ To evaluate the effects and possible mechanism of bFGF, EGF and suramin on the proliferation of cultured human meningioma cells in vitro.Methods\ Ten different primary early passage cultures derived from 10 patients with human meningioma underwent the intervention experiments by cell counting, MTT and DNA flow cytometry.Results\ Suramin significantly inhibited the growth of meningioma cells in culture. Meningioma cells were arrested in S and G2/M phases of the cell cycle.Growth factors (EGF and bFGF) induced cell proliferation was completely abolished by suramin.Conclusion\ Suramn acts as a scavenger neutralizing exogenous growth factors and affects the distribution of cell cycle.\;
