摘要
目的 研究正常人及生长激素分泌异常病人血清中胰岛素样生长因子结合蛋白3( I G F B P3) 的水平。方法 本文采用 Western 印迹方法测定20 例正常人,33 例活动性肢端肥大症病人和34 例特发性生长激素缺乏症( I G H D) 病人血清 I G F B P3 水平。结果 正常成人血清中存在五种分子量不同的 I G F B P, I G F B P3 含量为最高。本文测定了20 例正常成人和67 例生长激素分泌异常患者血清 I G F B P3 的相对光密度( R O D) ,正常人血清 I G F B P3 含量为(1 .1 ±0 .4) R O D,活动性肢端肥大病人为(2 .7 ±1 .2) R O D,明显高于正常成人( P< 0 .01) , I G H D 病人为(0 .4 ±0 .2) R O D,明显低于正常成人( P< 0 .01) 。血清生长激素与 I G F B P3 、胰岛素样生长因子 I与 I G F B P3 均呈正相关( P<0 .05) 。结论 Western 印迹测定血清 I G F B P3 正常值和病理值提示 I G F B P3 浓度的变化与生长激素功能状态密切相关,并依赖 G H。
Objective To investigate serum insulin like growth factor binding protein 3 (IGFBP 3) level in normal adults and patients with abnormality of growth hormone secretion. Methods Serum IGFBP 3 levels in 20 normal adults, 33 active acromegaly and 34 patients with idiopathic growth hormone deficiency (IGHD) were measured by Western blot. Results There are five kinds of IGFBPs with different molecular weight in serum of normal adult. IGFBP 3 was by far the major circulating binding protein. Serum IGFBP 3 level was found to be elevated in active acromegaly and diminished in IGHD. The relative optical density units (ROD) of IGFBP 3 were 1.1±0.4, 2.7±1.2 and 0.4±0.2 in normal adults, acromegaly and IGHD respectively. There was positive correlation between growth hormone and IGFBP 3 and between insulin like growth factor I and IGFBP 3. Conclusion The serum levels of IGFBP 3 measured by Western blot may reflect the growth hormone secreting status. Serum level of IGFBP 3 may provide a useful index for growth hormone function.
