摘要
目的:观察大黄酸对过氧化氢(H2O2)诱导损伤的血管内皮细胞增殖、凋亡、NO、NOS及SOD表达的影响,探讨大黄酸保护血管内皮细胞氧化损伤的可能机理。方法:100μmol/L H2O2造成血管内皮细胞氧化应激损伤模型,采用倒置显微镜观察细胞形态,Hochest染色观察细胞凋亡情况;MTT法和试剂盒测定各组细胞增殖、NO、NOS的及SOD的表达情况。结果:100μmol/LH2O2可损伤血管内皮细胞、抑制细胞增殖,促进细胞凋亡、降低NO、NOS含量和SOD的活力;而大黄酸则能改善H2O2诱导损伤的血管内皮细胞形态,促进其增殖,抑制其凋亡,提升NO、NOS的含量和SOD的活力,显示大黄酸对血管内皮细胞有一定的保护作用。
Objective: To observe the effect of the cell proliferation,apoptosis,NO,NOS and SOD expression of vascular endothelial cells injured by H2O2,the possible mechanism of rhein against apoptosis was discussed.Methods: Using the microscope and Hochest dyeing to observe the form of cells and apoptosis b;the method of MTT and kit were adopted to determine cell proliferation and expression of NO,NOS and SOD in each group.Result: 100μmol/L H2O2 can injury vascular endothelial cells and inhibit cell proliferation,promote cellular apoptosis and reduce NO,NOS content and SOD vitality;while rhein can improve form of vascular endothelial cell,promote cellular proliferation,restrain cellular apoptosis,advance NO,NOS content and SOD vitality by H2O2.Conclusion: Rhein has a protective effect of vascular endothelial cells.
