辛夷挥发油对变应性鼻炎大鼠IL-12、IFN-γ及组胺的影响 被引量:32

Effects of VOM bp on IL-12,IFN-γ and histamine levels of rats with allergic rhinitis

摘要 目的:研究辛夷挥发油(VOM bp)对实验性变应性鼻炎(AR)大鼠血清IL-12、IFN-γ及外周血组胺水平的影响,探讨IL-12、IFN-γ在AR发病机制中的作用及意义。方法:用卵清白蛋白(OVA)、氢氧化铝联合致敏建立变应性鼻炎大鼠模型,VOM bp连续灌胃给药5天,荧光法测外周血组胺水平,辅以行为学评分考察模型制备及药物治疗情况,并用酶联免疫吸附试验法(ELISA法)检测各组大鼠外周血清IL-12、IFN-γ的变化。结果:VOM bp(20~180mg/kg)能显著减轻AR大鼠鼻部过敏症状。VOM bp中(60 mg/kg)、低(20 mg/kg)剂量组与AR模型组比,能显著升高血清IL-12、IFN-γ,降低外周血组胺水平;VOM bp高剂量(180 mg/kg)组则能维持血清IL-12、IFN-γ及外周血组胺的正常水平,与正常组比无显著性差异。结论:IL-12和IFN-γ在AR中是主要炎性介质组胺释放的重要影响因素;VOM bp可通过影响IL-12、IFN-γ水平调节外周血组胺含量,减轻炎症反应。 Objective: To investigate the effect of Flos Magnoliae volatile oil(VOM bp) on IL-12,IFN-γ and histamine levels in blood of the experimental allergic rhinitis(AR) rats;and study the role of IL-12,IFN-γ in AR mechanism.Method: Using the model of allergic rhinitis in rats caused by ovalbumin(OVA) and Al(OH)3 VOM bp ig 5 days then observed the effect through the changes of ethology scores and histamine levels.Histamine levels were measured by fluorometric method.Enzyme linked immunosorbent assay(ELISA) was used to investigate the levels of IL-12,IFN-γ in serum.Results: VOM bp(20~180mg/kg) significantly relieved the ethology symptoms of allergic rhinitis in AR rats.Compare with AR group,low-VOM bp(20 mg/kg) and middle-VOM bp(60 mg/kg) groups significantly decreased histamine and increased IL-12,IFN-γ levels.Besides that,high-VOM bp(180 mg/kg) group can maintain the levels of IL-12,IFN-γ and histamine in normal,compare with normal group there were no significantly differences.Conclusion: IL-12 and IFN-γ are important factors in AR and can affect the major inflammation mediator histamine conspicuously.VOM bp can regulate the releasing of histamine and relieve inflammation symptoms by affect the levels of IL-12 and IFN-γ.
