康脑液2号对局灶性脑缺血再灌注大鼠的治疗作用及其机制

摘要 目的观察康脑液2号对局灶性脑缺血再灌注(I/R)大鼠的治疗作用。方法 150只雄性SD大鼠随机分为假手术组、模型组、康脑液2号高、中、低剂量组。线栓法制备大鼠右侧大脑中动脉闭塞(MCAO)模型。大鼠缺血2 h,于再灌注24 h后观察神经功能,再灌注1、3、7,14,21 d,HE染色观察病理变化,免疫组织化学法观察各组大鼠血管内皮生长因子(VEGF)、血管生成素-1(Ang-1)表达,再灌注21 d免疫组织化学法标记CD105检测血管密度。结果与模型组相比,康脑液2号治疗组神经功能及病理改变明显改善,各种促血管生成因子明显多于模型组(P<0.05或P<0.01),脑组织血管显著增多(P<0.05或P<0.01)。结论康脑液2号能治疗脑I/R大鼠,可能促进脑I/R大鼠缺血周围区域血管新生。 Objective To investigate the effects of No.2 kangnao liquid on rats with focal cerebral ischemia-reperfusion(I/R).Methods 150 healthy male SD rats were randomly divided into 5 groups:sham operated,model,high-,middle-and low-dose No.2 kangnao liquid groups.The animal models of middle cerebral artery occlusion(MCAO) were established by a monofilament method from right external-internal carotid artery.Neurological evaluation was performed 24 h after reperfusion,and immunohistochemical techniques were employed to test the protein expression of VEGF,Ang-1 and CD105.Pathological changes were detected by HE staining.Results Compared with model group,No.2 kangnao group had the significantly improved neurological responses,the increased expression of VEGF and Ang-1 and the higher number of blood vessels(P0.05 or P0.01).Conclusions No.2 kangnao liquid can treat I/R rats through promoting angiogenesis.
