不同年龄组大鼠抑郁症模型评价 被引量:10

摘要 目的观察年龄对抑郁模型大鼠行为及生化方面的影响。方法选取SD雄性大鼠,按照年龄分为幼年、成年和中年,采用慢性轻度不可预见性应激刺激与孤养相结合的方法建立抑郁症动物模型,运用开野法(Open-Field)与糖水消耗量评定大鼠行为学改变,用比色法测量大鼠海马区单胺氧化酶(monoamine oxidase,MAO)活性,用酶联免疫吸附法检测血清促肾上腺皮质激素释放激素(CRH)、促肾上腺皮质激素(ACTH)、皮质酮(CORT)含量。结果与对照组相比,各组模型组糖水消耗量与Open-Field得分均下降(P<0.05),成年抑郁组比幼年抑郁组均下降(P<0.05),中年抑郁组比成年抑郁组均下降(P<0.05);MAO活性与CRH、ACTH、CORT含量,模型组比对照组均升高(P<0.05),中年抑郁组比幼年抑郁组、成年抑郁组均升高(P<0.05),成年抑郁组与幼年抑郁组没有显著性变化(P>0.05)。结论刺激强度相等条件下,年龄的增长与抑郁模型大鼠行为学指标呈负相关,与生化指标呈正相关。
