摘要
建立了5种喹诺酮类药物的毛细管胶束电动色谱分离和尿样品的定量测定方法。测定条件为:运行缓冲液:50mmol/L磷酸二氢钠-100mmol/L溴化十六烷基三甲胺(1∶1,pH4.5);操作电压12KV,检测波长280nm。20min内5种喹诺酮药物全部得到分离,并与毛细管区带电泳作了比较。尿液对样品的测定无干扰。线性范围为5.0~35.0mg/L,最低检测浓度为2mg/L,RSD小于10%。
A method has been developed for the separation and determination of quinolones (pipemidic acid, lomefloxacin, levofloxacin, ciprofloxacin and norfloxacin) in human urine by micellar electrokinetic capillary chromatography (MEKC). Measurements were carried out in a buffer solution containing 25 mmol/L sodium dihydrogen phosphate and 50 mmol/L cetyltrimethylammonium bromide (CTAB), (pH4.5), at 12KV running voltage applied to a 50cm×50μm i.d. (45.5 cm effective length) capillary. Selectivity in the separation of five quinolones was improved by using MEKC in comparision with capillary zone electrophoresis (CZE). By MEKC, we determinated the concentrations of five quinolones in human urine. The recovery ranged from 85.07% to 113.70% with RSD less than 10%. The linear ranges were 5.0 ̄35.0mg/ml.
