摘要
目的探讨大鼠抑郁模型的血清细胞因子IL-2、IL-6及IL-10的动态变化及临床意义。方法 30只SD大鼠根据行为学评分后,随机分为对照组和抑郁组,每组15只。运用慢性不可预知应激及孤养模式建立抑郁模型,应激共8周。于建模开始前及每两周末进行大鼠体重测量、液体消耗实验及Morris水迷宫试验,并用酶联免疫吸附法(ELISA)检测每组大鼠的血清炎症因子IL-2、IL-6及IL-10的变化。结果各项行为学指标表明建模成功并呈动态变化。IL-2在0~4周时缓慢上升,至第4周时,抑郁组较对照组差异有显著性(P<0.05),其后缓慢下降;IL-6在第2周时,抑郁组就明显上升,较对照组差异有显著性(P<0.05),2~4周继续上升(P<0.01),至第6周时下降,但第8周时略上升;IL-10在第2周时,抑郁组略下降,第2~6周时上升,于第6周时较对照组差异有显著性(P<0.05),第8周时保持较高水平。结论大鼠抑郁模型的血清细胞因子IL-2、IL-6及IL-10存在异常,并随着应激的推进呈动态变化。
【Objective】 To explore the dynamic changes and significance of the serum cytokines(IL-2,IL-6,IL-10) of rat depressive model.【Methods】 Based on the evaluation of ethology,fifty SD rats were randomly and equally divided into two groups: the control group and the model group which was exposed to chronic unpredictable mild stress for 8 weeks.The body weight,fluid consumption test and Morris water maze test were measured while the cytokines(IL-2,IL-6,IL-10) in serum were done by ELISA at the beginning and per two weeks.【Results】 Based on the behavioral data,the rat depressive model has been established successfully,changing dynamically.IL-2 of the depressive group increased from the beginning to 4th week when it was significantly higher at 4th week(P 0.05) and then decreased slowly.IL-6 of the depressive group was significantly higher at the second week(P 0.05) and increased from 2nd to 4th week,while decreased at 6th week and increased a litter at 8th week.IL-10 of the depressive group decreased at 2nd week and increased from 4th to 6th week when it's significantly higher(P 0.05),and kept high level at 8th week.【Conclusion】 The serum cytokines(IL-2,IL-6,IL-10) of rat depressive model were abnormal,and changed dynamically with the development of stress.
