芹菜素对雌性大鼠卵巢颗粒细胞凋亡影响 被引量:1

Impact of apigenin on ovarian granulosa cell apoptosis in female rats

摘要 目的探讨芹菜素(apigenin,Api)对大鼠卵巢颗粒细胞凋亡的影响。方法选情前期成年雌性SD大鼠20只,取卵巢颗粒细胞进行原代细菌培养,贴壁后随机分为4组(对照组、Api 10-4M组、Api 10-5M组、Api 10-6M组),加入处理因素。培养结束后,收集各组培养卵巢颗粒细胞,经苏木精-伊红染色和吖啶橙染色观察颗粒细胞形态,经MTT法及流式细胞仪检测颗粒细胞凋亡率,观察Api对大鼠卵巢颗粒细胞凋亡的影响。结果 MTT法检测对照组,Api 10-4M组、Api 10-5M组、Api 10-6M组A值分别为(0.87±0.059)、(0.29±0.057)(0.49±0.03)、(0.63±0.035);实验组与对照组比较差异有统计学意义(P<0.01),提示实验组颗粒细胞数量减少,出现凋亡细胞;流式细胞仪检测对照组、Api 10-4M组、Api 10-5M组、Api 10-6M组的颗粒细胞凋亡率分别为(3.63±0.37)%、(36.37±2.28)%、(34.67±1.96)%、(30.92±1.65)%,实验组与对照组比较差异有统计学意义(P<0.01),提示实验组颗粒细胞凋亡率明显增高。结论 Api能抑制卵巢颗粒细胞的增殖,并促进其凋亡。 Objective To investigate the impact of apigenin(Api) on ovarian granulosa cell apoptosis in female rats.Methods Twenty adult female proestrus SD rats were selected and their granulosa cells were treated with primary cell culture.After adherence,the cells were randomly divided into four groups(control group,groups treated with 10-4M,10-5M,and 10-6M of Api).After the treatment,all ovarian granulosa cells were gathered and stained with heamatoxylin-eosin(HE) and acridine orange(AO)to observe the form of granulosa cell.Granulosa cell apoptosis rate was detected by methyl thiazolyl tetrazolium(MTT) assay and flow cytometry to observe the effect of Api on apoptosis of ovarian granulosa cell.Results The A values for the control,10-4M,10-5M,and 10-6M Api groups were 0.87±0.059,0.29±0.057,0.49±0.03,and 0.63±0.035,respectively,with significant differences between the treated and control group(P0.01 for all),indicating the decrease of granulosa cells and the increase of apoptosis.The rates of granulosa cell apoptosis for control,10-4M,10-5M,and 10-6M Api group were 3.63%±0.37%,36.37%±2.28%,34.67%±1.96%,and 30.92%±1.65%,respectively,with significant differences between the treated and control group(P0.01 for all),indicating the increase of granulosa cell apoptosis in experimental group.Conclusion Api can inhibit the proliferation and promote the apoptosis of ovarian granulosa cells.
